I also enjoy New York's antics from The Flavor of Love and her new show. Funny stuff.
House, definitely. Even if the show can be somewhat formulaic, Laurie's performance makes the show a must for me. Vic Mackey - A sociopath that you really, REALLY want on your side. Are we better or worse for having him on the streets? Michael Scott - Every awkward person I've ever met, summed up in one character. The best is when he actually comes through as charming - like when you see him make sales or in the most recent episode where he visited Pam at her art show. Dr. Zoidberg - A crustacean doctor with astonishingly low self-esteem. Genius. Tracy Jordan - Ridiculous and hilariously over the top. I have to say, 30 Rock is probably my new favorite show. GOB Bluth - He has illusions. Not tricks. Will Arnett's performance is legitimately genius work. Hiro Nakamura - The main reason to watch Heroes. He's just so charming and dedicated to his mission. Darnell/Crabman - The actor plays him as the most laconic yet intelligent person in the entire world of the show. Love it. Dr. Perry Cox - House before there was House. A supreme asshole who at least used to be one of the best characters in any show.
Late addition: Gaius Baltar - The way he's played makes him so evil and sympathetic. He's fantastic.

I think it's pretty tough to argue against Dr. Cox, who I've seen on a few lists. I agree with Chamberk that his character (along with Scrubs in general) has kind of fallen off a bit. Even so, he remains an extremely pertinant character.
I'll throw in Carl from Aqua Teen Hunger Force. Seems like any episode where he's a central part of the plot is hilarious.

One of my favorites right now is Ari Gold on Entourage. I've always been a fan of Jeremy Piven, but Ari is his masterpiece. Without him, there is no show.
