I think the best might be Bjork in "Dancer in the Dark", the Lars von Trier movie. She's great in it, unlike anything I've seen before. Unfortunately I had a few small issues with the movie itself, but nothing wrong with her acting at all. Madonna I really enjoyed in "A League of Their Own" and "Dick Tracy" And of course Frank Sinatra and Dean Martin movies. I think they were primarilly singers/crooners before they got into movies. I know they really shouldn't count though.

Personally I thought glen hansard and marketa irglova were spectacular in Once. Although I suppose it could be argued how much they were "acting" or w/e but regardless they were good.
I'd second A Hard Day's Night as fine acting by musicians.
Jason Schwartzman, has released an album under Coconut Records and has given some good performances in Rushmore and The Darjeeling Limited. He's also been in Phantom Planet.

He was great in Mystery Men. There are times when an actor can say a simple throw away line, so well that it gets jammed in your head forever. Such as: "How about that Defraculator thing, how'd that turn out?" as he's sitting down onto the couch. Also, I though Russel Crowe, lead singer for 30 Odd Foot of Grunts, was pretty good in the movie Gladiator.

Bjork - Dancer in the DarkHer performance knocked the shit out of me; I've only seen the movie three times, because I can't handle it--I cry so hard snot runs down my neck. EDIT: Just noticed it's already been mentioned.
